Introduction: Syphilis is one of the most common sexually transmitted diseases in the world. Although it is curable, there is a worrying increase in the number of cases of this pathology in the country. In Rondônia, the lack of local research, combined with the growth in cases, justifies the need for this study, which aims to analyze the prevalence, characteristics of the affected population, and transmission factors to contribute to reducing the associated risks. Objective: The objective was defined as establishing the epidemiological profile of acquired syphilis in a municipality in the interior of Rondônia, Vilhena. Methodology: The research method was based on an observational, cross-sectional, descriptive, and retrospective study, with a quantitative approach developed from the notification records through the Sistema de Informação de Agravos de Notificação (SINAN) database. Results: In total, of the 643 reported cases from 2018 to 2022, the majority were male, between 20 and 29 years old, with ignored or unfilled educational levels, and self-declared as mixed race. Conclusions: Thus, due to the increase in the number of syphilis cases in Vilhena, it is concluded that there is a need for prevention and screening actions for the disease, especially in populations considered vulnerable.
